目的研究长期应用大黄对结肠肌电皮动力基本功能单位的影响,探讨Cajal间质细胞(ICC)在脾虚大鼠结肠电中的作用。方法50只大鼠随机分为正常对照组、大黄组,3个月后测定结肠慢波频率及振幅,用碘化锌-锇酸法(ZIO)观察肌间神经丛及ICC变化,透射电镜观察结肠动力基本功能单位即壁内神经-Cajal间质细胞-平滑肌细胞(ENS-ICC-SMC)的病理改变。结果长期服用大黄所致脾虚大鼠,结肠慢波频率及振幅明显下降;结肠电节律失常;慢波频率变异系数(CV)及异常节律指数(ARI)显著增加;肌间丛神经纤维分布不均匀,突起连接杂乱,视野ICC稀少;电镜下见肌间神经轴突空化,ICC细胞样变性。结论长期服用大黄导致脾虚大鼠结肠慢波频率减慢,其机制可能为结肠动力基本功能单位的病理改变。
Objective To study the effect of long-term application of rhubarb on the basic functional units of colon muscle electromyocytes, and to explore the role of interstitial cells of Cajal (ICC) in the colonic electrical activity of rats with spleen deficiency. Methods 50 rats were randomly divided into normal control group and rhubarum group. The frequency and amplitude of slow wave in the colon were measured after 3 months. The changes of myenteric plexus and ICC were observed by zinc iodide-decanoic acid (ZIO) method. Transmission electron microscope observation The basic functional unit of colonic function is the pathological changes of intramural nerve-Cajal mesenchymal cell-smooth muscle cells (ENS-ICC-SMC). Results The long-term administration of rhubarb in rats with spleen deficiency, the frequency and amplitude of colonic slow waves decreased significantly; the electrical rhythm of the colon was abnormal; the slow wave frequency variation coefficient (CV) and aberrant rhythm index (ARI) increased significantly; the distribution of intermuscular plexus nerve fibers was not uniform , Prominent connection disorder, ICC scarcity; Electron microscopy see axon cavitation, ICC cell degeneration. Conclusion Long-term use of rhubarb caused slowing of the frequency of colonic slow waves in spleen-deficient rats. The mechanism may be the pathological changes of the basic functional units of colonic motility.
