北柴胡镇痛有效物质组提取纯化工艺研究

目的:北柴胡为临床镇痛方剂的组成药味之一,经前期文献调研及药效学实验,确定北柴胡镇痛的有效物质组为多糖类成分,实验以多糖含量和出膏率为指标,探讨北柴胡多糖组分的提取纯化工艺条件。方法:采用L9(34)正交试验,以多糖含量和出膏率的综合指标为考察指标,对影响提取工艺的提取温度、溶剂量、提取次数、提取时间进行考察;以多糖纯度为指标,对影响纯化工艺的浓缩指数、醇浓度、静置时间进行考察。结果:确定柴胡多糖组分最佳提取条件为提取温度100℃,15倍量水,回流提取3次,每次1h;最佳纯化条件为提取液浓缩成1g/mL,80%的乙醇沉淀,静置6h。结论:确定北柴胡镇痛有效物质组的最佳提取纯化工艺条件,该优化工艺条件合理、可靠,在此工艺下得到的多糖组分纯度为30.69%。 OBJECTIVE: Bupleurum chinense is one of the ingredients of clinical analgesic prescription. According to the literature review and pharmacodynamic experiments before, Bupleurum chinense was determined to be an effective substance group for polysaccharides. The content of polysaccharide and the rate of ointment As an indicator to explore the extraction and purification conditions of polysaccharides components of Bupleurum. Methods: The L9 (34) orthogonal test was used to evaluate the extraction temperature, solvent amount, extraction times and extraction time with the comprehensive index of polysaccharide content and the rate of ointment as the index. Concentration index affecting the purification process, alcohol concentration, resting time were investigated. Results: The optimal extraction conditions of polysaccharides from Radix Bupleuri were extraction temperature of 100 ℃ and 15 times amount of water, and refluxing for 3 times, each time for 1 hour. The optimal purification conditions were as follows: the extract was concentrated to 1 g / mL and 80% ethanol precipitation , Stand for 6h. Conclusion: The optimum extraction and purification conditions for the active substance of Bupleurum in Bupleurum chinense were determined. The optimized conditions were reasonable and reliable. The purity of the polysaccharide fraction was 30.69%.
