【目的】提取分离桂木叶中活性成分儿茶素并建立其含量测定方法。【方法】采用硅胶柱层析及重结晶法分离桂木叶中儿茶素,通过光谱和波谱分析鉴定其化学结构。采用高效液相色谱(HPLC)法进行儿茶素的含量测定,色谱条件:Phenomenex prodigy C18色谱柱(250 mm×4.60 mm,5μm),流动相:乙腈—体积分数0.4%磷酸水梯度洗脱,流速:1.2 m L/min,检测波长:280 nm,柱温35℃,进样量10μL。【结果】经光谱和波谱分析鉴定,从桂木叶中分离得到的化合物为儿茶素,其纯度≥980 mg/g;儿茶素在20.04~120.24μg/m L范围内与峰面积线性关系良好(r=0.999 8);不同批次桂木叶样品中儿茶素含量为0.58~1.85 mg/g,平均回收率为98.07%(sR=1.84%)。【结论】首次确认了桂木叶含活性成分儿茶素,并建立其HPLC含量测定方法,所建方法简便、快速、定量准确,可为桂木叶的质量标准及资源开发提供参考依据。
【Objective】 Extraction and separation of catechin in cinnamon leaves and determination of its content were established. 【Method】 Silica gel column chromatography and recrystallization were used to separate the catechins from the leaves of Cinnamomum cassiae. Their chemical structures were identified by spectroscopic and spectral analysis. The content of catechins was determined by high performance liquid chromatography (HPLC). The chromatographic conditions were as follows: Phenomenex prodigy C18 column (250 mm × 4.60 mm, 5 μm), mobile phase: gradient elution with 0.4% Flow rate: 1.2 m L / min, detection wavelength: 280 nm, column temperature 35 ℃, injection volume 10μL. 【Result】 The results showed that the compounds isolated from the leaves of Cinnamomum cassiae were catechin with the purity of ≥980 mg / g. The linear relationship between catechin and peak area was good in the range of 20.04 ~ 120.24 μg / m L (r = 0.999 8). The catechin content of different batches of cinnamon leaves was 0.58-1.85 mg / g with the average recovery of 98.07% (sR = 1.84%). 【Conclusion】 The cinnamic acid content in cinnamon leaves was confirmed for the first time, and the determination method of its HPLC content was established. The established method is simple, rapid and accurate. It can provide a reference for the quality standard and resource exploitation of cinnamon leaves.
