目的:研究六月青多糖(LYQP)对四氯化碳(carbon tetrachloride,CCl4)所致大鼠肝纤维化的干预作用及机制。方法:SD大鼠随机分成两组,模型组大鼠采用50%CCl4花生油溶液1 mL.kg-1ig造模,每周2次,连续7周,正常组用生理盐水代替。将病理检查确认肝纤维化已形成的SD大鼠60只随机分为LYQP(600,300,150 mg.kg-1)组,秋水仙碱(0.2 mg.kg-1)组,CCl4模型对照组,每组12只。于造模的第8周开始给药。正常组和CCl4模型对照组ig生理盐水,药物组ig LYQP和秋水仙碱,每日1次,连续ig给药4周。检测大鼠血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、谷胱甘肽-过氧化物酶(GSH-Px)活性以及肝组织中羟脯氨酸(Hyp)的含量,采用Masson染色观察肝组织纤维化程度。结果:LYQP(600,300,150 mg.kg-1)组ALT和AST活性为(13.53±2.30),(19.89±2.13),(26.31±2.87)U.mL-1,(17.14±2.96),(24.85±3.14),(33.63±3.91)U.mL-1,与模型对照组比较,LYQP可明显降低CCl4致大鼠纤维化血清ALT和AST活性;GSH-Px活性为(710.4±35.98),(653.8±25.36),(612.0±27.96)U.mg-1,Hyp含量为(0.382±0.017),(0.446±0.022),(0.517±0.024)mg.g-1与模型对照组比较,LYQP可明显增高GSH-Px活性和降低Hyp含量;Masson染色显示各药物组干预的大鼠肝脏胶原纤维均有所减少。结论:六月青多糖可减轻CCl4诱导的肝纤维化。
Objective: To investigate the intervention effect and mechanism of LYQP on liver fibrosis induced by carbon tetrachloride (CCl4) in rats. Methods: SD rats were randomly divided into two groups. Rats in model group were treated with 1 mL.kg-1ig of 50% CCl4 peanut oil solution twice a week for 7 weeks. Normal rats were replaced by normal saline. Sixty Sprague-Dawley rats were randomly divided into LYQP group (600, 300 and 150 mg.kg-1), colchicine group (0.2 mg.kg-1) and CCl4 model group only. The first 8 weeks of model administration. Normal group and CCl4 model control group ig saline, drug group ig LYQP and colchicine, once a day, continuous ig administration for 4 weeks. The levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), glutathione peroxidase (GSH-Px) and hydroxyproline (Hyp) Masson staining was used to observe the degree of hepatic fibrosis. RESULTS: The ALT and AST activities of LYQP group were (13.53 ± 2.30), (19.89 ± 2.13), (26.31 ± 2.87) U.mL-1, (17.14 ± 2.96), (24.85 ± 3.14) ) And (33.63 ± 3.91) U.mL-1, respectively. Compared with the model control group, LYQP could significantly decrease the ALT and AST activities of fibrosis serum induced by CCl4. The activity of GSH-Px was (710.4 ± 35.98), (653.8 ± 25.36 ), (612.0 ± 27.96) U.mg-1 and Hyp contents were 0.382 ± 0.017, 0.446 ± 0.022 and 0.517 ± 0.024 mg.g-1, respectively. Compared with the model control group, LYQP could significantly increase GSH- Px activity and decreased Hyp content; Masson staining showed that the hepatic collagen fibers of rats in each drug group decreased. Conclusion: Junqing polysaccharide can reduce CCl4-induced liver fibrosis.
