目的应用同位素标记相对与绝对定量技术(iTRAQ)分析先天性卵巢发育不良综合征(Turner综合征)患者外周血单核细胞的总蛋白,并找出蛋白质组的表达差异。方法选取2014年4月-2015年10月在深圳市人民医院确诊的Turner综合征患者5例(试验组),另选取5名健康女性作为健康对照组。采用强阳离子交换色谱分析法的8种不同同位素标记的iTRAQ和液相色谱法以及串联质谱分析Turner综合征患者和健康对照组女性外周血单核细胞的总蛋白,应用Mascot软件分析和基因本体论(GO)分析表达差异的蛋白。结果共有25个差异表达蛋白质被确定,其中包括17个蛋白表达上调和8个蛋白表达下调。这些表达差异的蛋白中选择3个显著表达差异作为候选蛋白,分别是表达上调的性激素结合球蛋白、甲状腺素结合球蛋白及表达下调的甲状旁腺激素原。结论性激素结合球蛋白、甲状腺素结合球蛋白及甲状旁腺激素原等显著差异蛋白可能作为该病潜在的临床筛查生物标志物。
Objective To analyze the total protein of peripheral blood mononuclear cells in patients with congenital ovarian dysplasia syndrome (Turner syndrome) by using isotope labeled relative and absolute quantification (iTRAQ) and to find out the difference of proteome expression. Methods Five patients (experimental group) with Turner syndrome diagnosed in Shenzhen People’s Hospital from April 2014 to October 2015 were selected. Five healthy women were selected as healthy control group. 8 different isotope-labeled iTRAQ and LC-MS were analyzed by strong cation exchange chromatography. Total protein in peripheral blood mononuclear cells of Turner syndrome patients and healthy controls were analyzed using Mascot software and gene ontology (GO) to analyze differentially expressed proteins. Results A total of 25 differentially expressed proteins were identified, including 17 proteins up-regulated and 8 down-regulated. Three of these differentially expressed proteins were selected as candidate proteins, which are upregulated sex hormone binding globulin, thyroxine binding globulin and downregulated parathyroid hormone. Conclusions Significant differential proteins such as sex hormone binding globulin, thyroxine binding globulin and parathyroid hormone may serve as potential clinical screening biomarkers for the disease.
