目的观察壬基酚(NP)对大鼠血浆和尿液中5-羟色胺(5-hydroxy tryptamine,5-HT)及血小板中5-HT和5-HT2A受体含量的影响,探讨壬基酚对大鼠5-HT和5-HT2A受体影响的毒效应机制。方法将24只SD雄性大鼠分为阴性对照组和NP低、中、高剂量组[30、90、270mg/(kg·d)],隔日灌胃染毒28d后检测大鼠血浆中5-HT、血小板中5-HT和5-HT2A受体含量,并检测灌胃后收集到的24h尿液中5-HT的含量。结果染毒28d后,随NP暴露的剂量增加,各组大鼠血浆、血小板及尿液中5-HT含量升高,血小板中5-HT2A受体表达则下降。NP暴露中、高剂量组大鼠血浆及血小板中5-HT含量均高于对照组(P<0.01;P<0.01),NP暴露高剂量组大鼠血小板5-HT2A受体表达低于对照组(P<0.01)。第4~28天,NP暴露低、中、高剂量组大鼠的尿液5-HT含量均高于对照组(P<0.01)。结论 NP暴露剂量与大鼠血小板5-HT、5-HT2A受体及血浆和尿液中5-HT含量均呈剂量-效应关系,提示NP通过影响大鼠5-HT水平和5-HT2A受体表达而产生毒效应。
Objective To observe the effects of nonylphenol (NP) on 5-hydroxytryptamine (5-HT) and platelet 5-HT and 5-HT2A receptor in rat plasma and urine, Toxicity Mechanism of 5-HT and 5-HT2A Receptors in Rats. Methods Twenty-four SD male rats were divided into negative control group and NP low, medium and high dose groups [30, 90, 270mg / (kg · d)], and the rats were intragastrically administered for 28 days. HT, 5-HT and 5-HT2A receptors in platelets, and detect the contents of 5-HT in 24h urine collected after intragastric administration. Results After 28 days of exposure, the levels of 5-HT in plasma, platelet and urine increased and the expression of 5-HT2A receptor in platelets decreased with the dose of NP exposure. The levels of 5-HT in plasma and platelet of NP-exposed rats were higher than those of the control group (P <0.01; P <0.01). The expression of platelet 5-HT2A receptor in high-dose NP exposure group was lower than that of the control group (P <0.01). From the 4th to the 28th day, the urinary 5-HT contents of the rats in low, medium and high dose NP exposure groups were higher than those in the control group (P <0.01). Conclusion NP exposure dose-effect relationship with 5-HT, 5-HT2A receptor in rat platelets and 5-HT content in plasma and urine, suggesting that NP by affecting rat 5-HT levels and 5-HT2A receptor Expression and produce toxic effects.
