目的研究钙黏附分子E-cadherin(E-cad)、CD44变异体6(CD44v6)和基质金属蛋白酶(MMP2)在膀胱尿路上皮癌(bladder urothelial carcinoma)中的表达及对预后的意义。方法应用免疫组化Envision法,检测105例膀胱尿路上皮癌患者和20例正常体检者膀胱黏膜E-cad、CD44v6及MMP2的表达情况。结果 20例正常膀胱黏膜中E-cad、CD44v6均正常表达,MMP2均阴性表达,105例膀胱尿路上皮癌中E-cad及CD44v6的异常表达率分别为43.8%(46/105)和58.1%(61/105),MMP2阳性表达为53.3%(56/105)。E-cad、CD44v6的异常表达率和MMP2的阳性表达率与患者肿瘤的病理分级、临床分期及生存率相关。结论E-cad、CD44v6的异常表达和MMP2的阳性表达在膀胱尿路上皮癌的演变过程中起着重要作用,可作为监测肿瘤恶性程度和预后有效的分子生物学指标之一。
Objective To study the expression of E-cadherin, CD44v6 and MMP2 in bladder urothelial carcinoma and its prognostic significance. Methods The expression of E-cad, CD44v6 and MMP2 in bladder mucosa of 105 cases of bladder urothelial carcinoma and 20 cases of normal subjects were detected by immunohistochemistry Envision method. Results The positive expression rates of E-cad and CD44v6 in 20 cases of normal bladder mucosa were all negatively expressed in MMP2. The abnormal expression rates of E-cad and CD44v6 in 105 cases of bladder urothelial carcinoma were 43.8% (46/105) and 58.1% (61/105). The positive expression of MMP2 was 53.3% (56/105). The abnormal expression rates of E-cad and CD44v6 and the positive expression rate of MMP2 correlated with the pathological grade, clinical stage and survival rate of the patients. Conclusions The abnormal expression of E-cad and CD44v6 and the positive expression of MMP2 play an important role in the evolution of bladder urothelial carcinoma and can be used as one of the molecular biological indicators to monitor the malignant degree and prognosis of the tumor.
