以山鸡椒无菌苗叶片、茎段为外植体诱导愈伤组织,通过愈伤组织分化诱导不定芽途径,建立山鸡椒植株再生体系。结果表明,山鸡椒茎段是诱导愈伤组织最佳外植体,愈伤组织诱导和不定芽分化的最适培养基为MS+2.0 mg·L-1BA+0.1 mg·L-1IBA,分化培养基中添加适当浓度的维生素C可减轻愈伤组织褐化程度;诱导不定芽生根较理想的培养基为1/2MS+0.2 mg·L-1IBA+0.4 mg·L-1NAA。同时,通过组织学观察了愈伤组织的结构和不定芽的发生。
The stems and leaves of the pepper seedlings were used as explants to induce the callus, and the shoot regeneration system was established through inducing the adventitious buds through callus differentiation. The results showed that the optimal medium for the induction of callus and differentiation of adventitious buds was the optimum medium for the induction of callus by stem segments of B. chinense. MS medium supplemented with 2.0 mg · L-1 BA and 0.1 mg · L-1 IBA The appropriate concentration of vitamin C in the medium can reduce the degree of callus browning; the optimal culture medium for inducing adventitious buds is 1 / 2MS + 0.2 mg · L-1IBA + 0.4 mg · L-1 NAA. At the same time, the structure of callus and adventitious buds were observed through histology.
