[Objective] To establish a qualitative and quantitative analysis method for the flavonoids in HERB OF AIR-PLANT and preliminarily evaluate its antioxidant activity.[Method] After hydrolysis,the total flavonoids were extracted and determined by UV spectrophotometry.TLC was applied to detect quercetin and kaempferol.Fention method was applied to determine the antioxidant activity.And the total quercetin and kaempferol content in the flavonoids was determined by HPLC.[Result] Rutin showed good linear relationship in the range of 11.0-110.0 μg/ml(r=0.999 8) with the average recovery of 99.3%(n=6,RSD=1.3%) by the UV spectrophotometry.Quercetin and kaempferol were detected by TLC.Quercetin showed good linear relationship in the range of 37.5 to 1 200.0 ng/ml(r=0.999 7) with the average recovery rate of 101.67%(n=6,RSD=1.92%);and kaempferol showed good linear relationship in the range of 62.5 to 2 000.0 ng/ml(r=0.999 4) with the average recovery rate of 98.68%(n=6,RSD=1.40%).The total flavonoids extracted from HERB OF AIR-PLANT showed high antioxidant activity,and antioxidant effect of total flavonoids from HERB OF AIR-PLANT increased as the concentration of total flavonoids enhanced.[Conclusion] This method is accurate and reliable,and could be used for the qualitative and quantitative determination of the flavonoids and for the quality control of HERB OF AIR-PLANT.
