目的探讨外源性 p14ARF 基因对胰腺癌 PC-3细胞血管生成的影响。方法将转染了外源性 p14ARF 基因的人胰腺癌 PC-3细胞株接种裸鼠形成实体瘤,用 RT-PCR 和 Western blot 法观察 VEGFmRNA 及 VEGF 蛋白的表达,并对实体肿瘤的微血管密度进行检测。结果转染p14ARF 基因后的 PC-3细胞实体瘤中 VEGFmRNA 及 VEGF 蛋白的表达下调,血管密度降低。结论p14ARF 基因能下调内源性 VEGF 的表达,有着抗血管生成的治疗意义。
Objective To investigate the effect of exogenous p14ARF gene on the angiogenesis of pancreatic cancer PC-3 cells. Methods Human pancreatic cancer cell line PC-3 transfected with exogenous p14ARF gene was inoculated into nude mice to form solid tumors. The expression of VEGFmRNA and VEGF protein were observed by RT-PCR and Western blot. The microvessel density Detection. Results The expression of VEGF mRNA and VEGF protein in PC-3 cells after transfection of p14ARF gene was down-regulated and the density of blood vessels decreased. Conclusion The p14ARF gene can downregulate the expression of endogenous VEGF and has anti-angiogenic therapeutic effect.
