目的:探讨佐剂性关节炎(AA)大鼠滑膜细胞的类肿瘤样增生和相关基因表达的机制。方法:将Wistar大鼠随机分两组,每组10只,并应用弗氏完全佐剂诱发大鼠AA实验性模型,定期观测踝关节肿胀度至第40天。分别取AA模型组和正常对照组大鼠踝关节进行组织切片,用HE染色后观察病理学变化。取大鼠膝关节滑膜组织体外进行原代细胞培养,用MTT比色法检测大鼠滑膜细胞的增生。同时采用RTPCR法,检测模型组及正常对照组大鼠滑膜组织中相关基因Cmyc和ODCmRNA的转录水平。结果:①体外培养的AA模型组大鼠关节滑膜细胞的增殖显著高于正常组大鼠(P<0.01),并呈类肿瘤样增生。②AA模型组大鼠关节滑膜组织中,Cmyc和ODCmRNA的转录水平明显高于正常对照大鼠(P<0.01)。结论:AA模型组大鼠关节滑膜细胞的类肿瘤样增生,可能与其相关基因Cmyc和ODCmRNA的转录水平增高有关。
Objective: To investigate the mechanism of tumor-like hyperplasia and related gene expression in synovial cells of adjuvant arthritis (AA) rats. Methods: Wistar rats were randomly divided into two groups (n = 10 in each group). Experimental rats were induced by complete Freund’s adjuvant. The ankle swelling degree was observed regularly until day 40. The ankle joints of AA model group and normal control group were respectively taken for histological section. The pathological changes were observed with HE staining. The synovial tissues of the knee joints of rats were cultured in primary culture in vitro. The proliferation of synovial cells was detected by MTT assay. At the same time, RTPCR method was used to detect the transcription level of Cmyc and ODCmRNA in synovial tissue of model group and normal control group. Results: ① The proliferation of synovial cells in AA model group was significantly higher than that in normal group (P <0.01), and it was tumor-like hyperplasia. ② The transcription level of Cmyc and ODCmRNA in synovial tissue of AA model group was significantly higher than that of normal control group (P <0.01). CONCLUSION: The tumor-like hyperplasia of synovial cells in AA model group may be related to the increased transcription of Cmyc and ODCmRNA.
