目的探究羊膜泪道支架对雄兔干眼症的预防效果。方法选取36只健康雄兔,剪除第三眼睑并抗感染处理,随机分为3组(每组12只),制作去势雄兔模型,其中A组为阴性对照组,B组为干眼模型组,C组为泪道羊膜预防组,并在植入羊膜泪道支架前、植入后2周、4周及6周行角膜荧光素(fluorescent,FL)染色、Schirmer I试验、Western blot、免疫荧光染色及角膜共聚焦显微镜检查。结果三组不同时间点间的泪液分泌量及FL有明显改变(F=7.126,P=0.009;F=9.658,P=0.016),三组间泪液分泌量及FL有明显改变(F=12.582,P=0.005;F=13.187,P=0.013),三组的泪液分泌量及FL变化趋势也有明显改变(F=8.531,P=0.007;F=10.652,P=0.019)。A、B、C三组植入后6周上皮基底细胞分别(3811±414)个细胞、(3820±314)个细胞、(2789±353)个细胞,炎症细胞密度分别为(266±28)个细胞、(266±29)个细胞、(67±13)个细胞;A、B、C三组植入后6周上皮基底细胞(F=13.442,P=0.012)和炎症细胞密度(F=9.231,P=0.021)比较,差异均有统计学意义。泪道上皮K16染色均为阴性,植入羊膜泪道支架后各时间点A、B、C三组兔泪小管组织中α-SMA表达无明显改变,差异有统计学意义(F=14.681,P=0.002)。结论植入羊膜泪道支架对雄兔干眼症的预防有一定的效果。
Objective To investigate the preventive effect of amniotic lacrimal duct stent on dry eye of male rabbits. Methods Thirty-six healthy male rabbits were selected and the third eyelid was cut off for anti-infection treatment. The rabbits were randomly divided into 3 groups (12 in each group) to make castrated male rabbits model, in which group A was negative control group and group B was dry eye model Group C was lacrimal duct amniotic prophylaxis group. Corneal fluorescein (FL) staining, Schirmer I test and Western blot were performed at 2 weeks, 4 weeks and 6 weeks after laminectomy. Immunofluorescence staining and Confocal microscopy. Results There was a significant change in the amount of tear fluid and FL in three groups at different time points (F = 7.126, P = 0.009; F = 9.658, P = 0.016) P = 0.005; F = 13.187, P = 0.013). There was also a significant change in tear volume and FL in the three groups (F = 8.531, P = 0.007; F = 10.652, P = 0.019). The number of epithelial basal cells (3811 ± 414), (3820 ± 314) cells and (2789 ± 353) cells in group A, B and C at 6 weeks after implantation were (266 ± 28) (F = 13.442, P = 0.012) and inflammatory cell density at 6 weeks after implantation in group A, B, and C (F = 9.231, P = 0.021), the differences were statistically significant. Laryngeal epithelium K16 staining were negative, amniotic lacrimal duct scaffold after implantation at each time point A, B, C groups of rabbit lacrimal ductal tissue expression of α-SMA had no significant change, the difference was statistically significant (F = 14.681, P = 0.002). Conclusion The implantation of amniotic lacrimal support has some effect on the prevention of dry eye in male rabbits.
