应用原位分子杂交技术及细胞图象分析证实了人肝癌细胞株SMMC7721细胞经10μg/ml的GM_3/GD_3处理后,GD_3处理组细胞内N-ras基因mRNA的阳性信号弥漫分布于整个细胞,而经GM_3处理组和对照组阳性信号集中于核膜。GM_3/GD_3处理组及对照组c-fos基因mRNA的阳性信号均集中于核膜,但GM_3处理组信号强度最大。上述结果提示,GD_3可使某些肿瘤细胞向更加恶性的方向发展,而GM_3则可使某些肿瘤细胞向正常方向转化。
The in situ molecular hybridization technique and cell image analysis confirmed that after treatment of human hepatocellular carcinoma cell line SMMC7721 with 10 μg/ml GM_3/GD_3, the positive signal of N-ras mRNA in the GD_3 treated group diffusely distributed throughout the cell. Positive signals were concentrated in the nuclear membrane in the GM3 treated and control groups. The positive signals of c-fos mRNA in GM_3/GD_3 treatment group and control group were all concentrated in the nuclear membrane, but the signal intensity of GM_3 treatment group was the highest. The above results suggest that GD_3 can make certain tumor cells develop in a more malignant direction, while GM_3 can make certain tumor cells transform in a normal direction.
