研究小鼠肝脏枯否细胞(Kupffer cell,KC)的细胞特征和抗肿瘤功能。方法:用光学和扫描电子 显微镜观察KC的特征,用细胞毒性试验检测抗肿瘤功能。结果:KC具有典型的巨噬细胞的特征,细胞表面的 IgG Fc受体和补体C(3b)受体活性,以及吞噬功能都很强。正常昆明种小鼠肝脏分离的KC对同种肿瘤细胞介导低 水平的细胞毒性作用,并产生肿瘤坏死因子(TNF)。分别用BCG、双歧杆菌或乳杆菌在原位激活后,肝脏KC对相 同的肿瘤细胞介导的细胞毒性作用及产生TNF均明显增强。结论:KC对控制肝脏肿瘤细胞生长起重要作用。
To study the cell characteristics and antitumor function of Kupffer cell (KC) in mouse liver. Methods: The characteristics of KC were observed by optical and scanning electron microscopy, and cytotoxicity assays were used to detect anti-tumor function. RESULTS: KC was characteristic of a typical macrophage, and its cell surface IgG Fc receptor and complement C (3b) receptor activity, as well as phagocytosis were strong. The KC isolated from the liver of normal Kunming mice mediated low-level cytotoxicity against the same tumor cells and produced tumor necrosis factor (TNF). After activated in situ with BCG, Bifidobacterium or Lactobacillus, respectively, the liver KC significantly enhanced the same tumor cell-mediated cytotoxicity and TNF production. Conclusion: KC plays an important role in controlling the growth of liver tumor cells.
