目的探讨四川省目前流行的麻疹病毒的基因型别和特征。方法采用Vero/SLAM细胞分离培养麻疹病毒,通过逆转录-聚合酶链反应(Reverse Transcription-Polymerase Chain Reaction,RT-PCR)方法扩增麻疹病毒N基因羧基末端456 bp片段,并对PCR产物进行序列测定和同源性分析。结果 2011年四川省分离到10株麻疹病毒均为H1a基因亚型,与H1基因型代表毒株的同源性为97.1%~98.9%。结论 H1a基因亚型仍为四川省本土流行病毒株的优势亚型,近年来四川省流行的麻疹病毒无较大变异,优势病毒的持续传播与麻疹的发病有密切联系。
Objective To explore the genotypes and characteristics of the measles virus currently prevailing in Sichuan Province. Methods The measles virus was isolated and cultured in Vero / SLAM cells. The 456 bp fragment of the carboxyl terminus of the N gene of measles virus was amplified by reverse transcriptase-polymerase chain reaction (RT-PCR). The PCR products were sequenced Assay and Homology Analysis. Results In 2011, 10 isolates of measles virus in Sichuan Province were all subtypes H1a, which shared 97.1% -98.9% homology with H1 strain. Conclusion H1a subtype is still the dominant subtype of the endemic strains in Sichuan Province. In recent years, there is no significant variation in the prevalence of measles virus in Sichuan Province. The continuous transmission of dominant virus is closely related to the incidence of measles.
