应用末端脱氧核苷酸转移酶介导的dUTP原位切口末端标记 (TUNEL)检测技术和显微图像定量分析方法研究了悬浮培养中南方红豆杉 (Taxuschinensisvar.mairei)细胞团不同区域细胞凋亡的发生 ,并比较了不同区域的细胞形态。结果显示细胞团可以划分为三个不同的区域 :核心区的细胞结构已消失 ,可见一些分化良好的管状结构 ;中层区是由增殖能力旺盛的分生组织细胞所构成 ;外层区是由分化成熟的细胞所组成。细胞凋亡主要发生在细胞团分化成熟的外层区。以上结果说明细胞凋亡可能是分化终结的一种形式
Endothelial deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) and microscopic image quantitative analysis were used to study the apoptosis of Taxus chinensis var. Mairei cell clusters in suspension culture Occurred, and compared the cell morphology in different regions. The results showed that the cell mass can be divided into three different regions: the cellular structure of the core region has disappeared, showing some well-differentiated tubular structure; the middle region is composed of proliferating meristematic cells; the outer region is differentiated Mature cells. Apoptosis mainly occurs in the maturation of the outer cell mass. The above results suggest that apoptosis may be a form of terminal differentiation
