为了应用放射免疫测定法和免疫组织化学方法进行脑啡肽的研究,我们制备了抗亮氨酸脑啡肽血清(ALS)和抗甲硫氨酸脑啡肽(AMS)。主要是分别合成两种脑啡肽免疫原,用大白兔进行免疫,经一次强化后分离和制备抗血清,所得抗血清具有满意的灵敏度和精密度。其最小检出量ALS为25pg,AMS为60pg。并具有较好特异性;LEK对MEK的交叉反应率为0.44%,MEK对LEK为2.8%。用效点系统评分法,其结果ALS为30分,AMS为27分,均属较优质的抗血清,能满足于脑啡肽的放射免疫测定要求。
In order to study enkephalins using radioimmunoassay and immunohistochemistry methods, we prepared anti-leucine enkephalin serum (ALS) and anti-methionine enkephalin (AMS). The two enkephalin immunogens were synthesized respectively and immunized with white rabbits. After one time of fortification, the antisera were separated and prepared. The obtained antisera had satisfactory sensitivity and precision. The minimum detectable ALS was 25pg and AMS was 60pg. And had better specificity. The cross-reactivity rate of LEK to MEK was 0.44% and MEK to LEK was 2.8%. With the effective point system scoring method, the results of ALS was 30 points, AMS was 27 points, are better anti-serum, to meet the enkephalin radioimmunoassay requirements.
