目的检测并比较感染性心内膜炎中VCAM-1在左心和右心室肌肉和瓣膜组织中的mRNA表达水平和作用。方法7只健康兔对照,26只兔分别行二尖瓣、三尖瓣手术和经静脉注射金黄色葡萄球菌,术后24 h提取左右心室肌肉和瓣膜组织RNA,应用RT-PCR技术测量VCAM-1 mRNA表达水平,进行比较分析。结果金葡菌菌血症未引起心脏各部位VCAM-1 mRNA表达水平的明显变化;二尖瓣手术和三尖瓣手术后,右心室和三尖瓣表达水平均显著提高(P<0.05),显著高于二尖瓣和左心室(P<0.05);二尖瓣手术后左心室表达水平显著降低(P<0.05),三尖瓣手术后左心室表达水平有所提高,但差异无显著意义(P>0.05)。结论心脏左心和右心系统VCAM-1在感染性心内膜炎早期有着不均衡的分布和变化,并可能起保护作用。
Objective To detect and compare the mRNA expression of VCAM-1 in left and right ventricular muscles and valve tissues in infective endocarditis. Methods Seven healthy rabbits and 26 rabbits were given mitral and tricuspid valve surgery and intravenous injection of Staphylococcus aureus respectively. The left and right ventricular muscle and valve tissue RNAs were extracted 24 hours after operation. The expression of VCAM- 1 mRNA expression levels, comparative analysis. Results Staphylococcus aureus bacteremia did not cause obvious changes of VCAM-1 mRNA expression in all parts of the heart. After mitral valve surgery and tricuspid valve surgery, the expression of right ventricle and tricuspid valve increased significantly (P <0.05) (P <0.05). The left ventricular expression was significantly decreased after mitral valve surgery (P <0.05), while the left ventricular expression was increased after tricuspid valve surgery, but the difference was not significant (P> 0.05). Conclusions VCAM-1 in the left and right hearts of the heart has unbalanced distribution and changes in the early stages of infective endocarditis and may play a protective role.
