目的观察db/db鼠糖尿病视网膜病变(diabetic retinopathy,DR)的成模情况。方法每月取尾尖毛细血管血测小鼠空腹血糖值及体质量一次,共7次。7个月后,取10只db/db鼠和6只db/m鼠(正常对照组)做闪光视网膜电图(Flash electroretinogram,F-ERG)检测。结果 db/db鼠的体质量明显上升;但血糖值上升趋势不明显,实验组97%小鼠的血糖值未达到DR小鼠模型的血糖值(11.1 mmol/L)。F-ERG统计结果表明,实验组a波和b波的潜伏时比对照组延长,差异有统计学意义(P<0.01);实验组b波的振幅与对照组相比,差异有统计学意义(P<0.01);实验组Ops总波及O2的振幅与对照组相比,差异有统计学意义(P<0.01)。结论本实验中db/db鼠的空腹血糖值虽大部分未达标,但其视网膜已出现DR的早期改变。造模标准不能仅依靠空腹血糖值,还应结合视网膜的病变情况。db/db鼠的成模标准有待进一步研究及完善。
Objective To observe the modeling of diabetic retinopathy (DR) in db / db mice. Methods Fasting blood glucose and body weight were taken from the tail capillaries every month for 7 times. Seven months later, 10 db / db mice and 6 db / m mice (normal control group) were examined by flash electroretinogram (F-ERG). Results The body weight of db / db mice increased significantly; however, the increase of blood glucose level was not obvious. The blood glucose level of 97% of the mice in the experimental group did not reach the blood glucose level of 11.1 mmol / L. The results of F-ERG showed that the latency of a-wave and b-wave in the experimental group was longer than that of the control group (P <0.01), and the amplitude of the b-wave in the experimental group was significantly higher than that of the control group (P <0.01). The amplitudes of Ops and O2 in the experimental group were significantly different from those in the control group (P <0.01). Conclusion Although most of the fasting blood glucose values of db / db mice were not up to standard in this experiment, the early changes of DR in the retina have appeared. Modeling standards can not rely on fasting blood glucose values, but also the combination of retinal lesions. db / db rat modeling standards to be further studied and improved.
