目的:观察大鼠脑出血后神经缺损症状和线粒体膜电位的改变,及中药制剂平肝熄风汤的干预作用。方法:用Ⅶ型胶原酶于脑内定位注射诱导大鼠脑出血模型。神经缺损积分评定神经功能;用荧光分光光度计测定线粒体悬液的荧光强度反映线粒体膜电位。结果:大鼠脑出血后迅速出现神经缺损症状,平肝熄风汤能明显降低神经缺损症状积分(4h:8.80分,1d:6.80分,3d:2.80分,7d:1.60分)。脑出血大鼠线粒体膜电位于术后4h显著下降,1d时膜电位下降达高峰,3,7d逐步恢复,仍低于正常组和假手术组(t=7.352P<0.05)。平肝熄风汤治疗组在脑出血后1,3,7d,线粒体膜电位均明显高于模型组(t=2.583P<0.05)。结论:大鼠脑出血后线粒体膜电位下降;平肝熄风汤能阻断膜电位下降,同时能减轻出血后神经损伤。
OBJECTIVE: To observe the changes of neurological deficits and mitochondrial membrane potential after intracerebral hemorrhage in rats and the intervention of Pinggan Xifeng Decoction, a traditional Chinese medicine preparation. METHODS: A model of intracerebral hemorrhage induced by injection of type VII collagenase in the brain was established. Neurological deficit scores were used to assess neurological function; fluorescence intensity of mitochondrial suspensions was measured by fluorescence spectrophotometer to reflect mitochondrial membrane potential. RESULTS: After the cerebral hemorrhage of rats, there was a rapid neurological deficit. Pinggan Xifeng Decoction significantly reduced the neurological deficit scores (4h: 8.80, 1d: 6.80, 3d: 2.80, and 7d: 1.60). The mitochondrial membrane in rats with intracerebral hemorrhage was significantly decreased at 4 h postoperatively. The membrane potential decreased to a peak at 1 d and gradually recovered at 3 and 7 d. It was still lower than that in the normal and sham groups (t=7.352 P<0.05). In the Pinggan Xifeng Decoction group, the mitochondrial membrane potential was significantly higher than that of the model group at 1, 3, and 7 days after cerebral hemorrhage (t=2.583P<0.05). Conclusion: The mitochondrial membrane potential decreased after intracerebral hemorrhage in rats. Pinggan Xifeng Decoction can block the decrease of membrane potential and reduce the neurological damage after hemorrhage.
