目的观察祛风止动方对抽动障碍大鼠行为学的影响。方法清洁级SD大鼠应用阿扑吗啡(APO)诱导抽动障碍模型。APO用药3周后,将造模成功的大鼠分为模型组、中药组、西药组各8只,另设正常组8只。正常组以0.9%NaCl溶液(10ml/kg)腹腔注射4周,模型组继续予APO(2mg/kg)腹腔注射4周,中药组在模型组处理基础上给予祛风止动方(18.4g/kg)灌胃,西药组在模型组处理基础上给予氟哌啶醇(1mg/kg)灌胃,1次/d,连续4周。对模型大鼠进行刻板行为和开野实验的研究,评价药物疗效。结果①刻板行为:模型组刻板行为评分均高于正常组、中药组和西药组,而且在给药15min后差异有统计学意义(P<0.05);②开野实验:模型组大鼠穿越格子数、理毛次数、直立次数、粪便粒数各项结果与其余各组相比差异均有统计学意义,中药组和西药组的穿越格子数和理毛次数相比差异有统计学意义(P<0.05)。结论祛风止动方可显著降低APO诱导抽动模型大鼠的活动水平,增强其适应力。
Objective To observe the influence of Qufeng Zhidong Fang on the behavior of rats with tic disorder. Methods A rat model of tetanus induced by apomorphine (APO) was applied to clean SD rats. Three weeks after the administration of APO, the successful rats were divided into model group, traditional Chinese medicine group and western medicine group each with 8 rats, and another 8 normal rats. In the normal group, 0.9% NaCl solution (10 ml/kg) was intraperitoneally injected for 4 weeks. The model group continued to be intraperitoneally injected with APO (2 mg/kg) for 4 weeks. The Chinese medicine group was given a hurricane stop method on the basis of model group treatment (18.4 g/ Kg) gavage, western medicine group given haloperidol (1mg/kg) gavage on the basis of model group treatment, once / d, for 4 weeks. The model rats were subjected to stereotyped behavior and open field experiments to evaluate drug efficacy. RESULTS 1 Stereotype behavior: The stereotyped scores of the model group were higher than those of the normal group, the TCM group, and the western medicine group, and there was a significant difference after 15 min (P<0.05); 2 Open field experiment: the model group rats crossed the grid The differences in number, number of hairs, number of erections, and number of stool pellets were statistically significant compared with the other groups. There was a statistically significant difference between the Chinese medicine group and the western medicine group in the number of crossing trellis and the number of grooming (P). <0.05). Conclusion Qufeng stop can significantly reduce the activity level of APO-induced twitch rats and enhance their adaptability.
