目的 为了研究胚胎肾及肾癌中性糖脂的表达情况。方法 采用改良的 Hakomori的方法 ,提取了肾透明细胞癌及胚胎肾组织的中性糖脂 (neutral glycosphingolipids,N-GSL s) ,并进行了高效薄层层析。结果 胚胎肾 N- GSL s的表达随着胚胎的成熟而呈规律性的变化 ,肾癌 N- GSL s的表达存在着异质性 ,与胚胎期肾组织 N- GSL s表达相似 ,它们都不含有正常成人肾所具有的长糖链的红细胞糖苷脂(Globoside)。结论 中性糖脂参与了胚胎肾组织的成熟与分化过程 ,中性糖脂中长糖链红细胞糖苷脂与肾组织的恶变可能有关。
Objective To study the expression of neutral glycolipids in embryonic kidney and renal cell carcinoma. Methods Neutral glycosphingolipids (N-GSLs) were extracted from renal clear cell carcinoma and embryonic kidney using a modified Hakomori method. High performance thin layer chromatography (TLC) was performed. Results The expression of N-GSL in embryonic kidney changed regularly with the maturation of embryos. The expression of N-GSL s in renal cell carcinoma was heterogeneous, which was similar to the expression of N-GSL in embryonic kidney Globoside, which contains the long sugar chain of normal adult kidneys. Conclusion Neutral glycolipid is involved in the maturation and differentiation of embryonic kidney tissue. The glycation of glycoleiphenolide, a long glycan chain, may be related to the malignancy of renal tissues.
