目的 通过观察胰岛素样生长因子及其受体 (IGFs)和增殖细胞核抗原 (PCNA)在不同肝组织的表达 ,探讨它们与HBV -DNA整合在肝癌发生中的关系 ,为肝癌的发病机制和临床诊疗提供一定的理论依据。方法 肝癌和癌旁肝组织各 32例 ,慢性活动性乙型肝炎 12例 ,胎肝 10例和正常肝组织 8例 ,Southern杂交检测各种肝组织中HBV -DNA整合情况 ,胶体金免疫电镜技术观察不同肝组织中IGFs和PCNA在超微结构水平的分布与表达。结果 81 3% (2 6/32 )的肝癌和癌旁肝组织为HBV -DNA整合阳性。 83 3% (10 /12 )的慢性活动性乙型肝炎为HBV -DNA整合阳性。所有胎肝组织和正常成人肝组织均无HBV -DNA整合。 87 5 % (2 8/32 )的肝癌和84 4 % (2 7/32 )癌旁肝组织的表达PCNA ;肝癌组织表达胰岛素样生长因子Ⅰ (IGF -Ⅰ )和IFG -Ⅰ受体 (IGF -ⅠR)的阳性率分别为 4 0 6% (13/32 )和 5 6 3% (18/32 ) ;癌旁肝组织表达IGF -Ⅰ和IGF -ⅠR的阳性率分别为5 0 0 % (16/32 )和 65 6% (2 1/32 )。肝癌组织表达胰岛素样生长因子Ⅱ (IGF -Ⅱ )和IGF -Ⅱ受体 (IGF -ⅡR)的阳性率均为 78 1% (2 5 /32 ) ;癌旁肝组织表达IGF -Ⅱ和IGF -ⅡR的阳性率均为 81 3% (2 6/32 )。HBV -DNA整合组中 ,肝癌和癌旁肝组织表达IGF -Ⅱ和IGF -ⅡR的阳性率?
Objective To observe the expression of insulin-like growth factor (IGF) and its receptors (IGFs) and proliferating cell nuclear antigen (PCNA) in different liver tissues, and to explore the relationship between their integration with HBV-DNA in the occurrence of hepatocellular carcinoma and the pathogenesis and clinical diagnosis and treatment of liver cancer. Methods 32 cases of hepatocellular carcinoma and adjacent liver tissues, 12 cases of chronic active hepatitis B, 10 cases of fetal liver and 8 cases of normal liver tissues. There was no HBV-DNA integration in all fetal liver tissue and normal adult liver tissue. The expression of PCNA was detected in 87.5% (2 8/32) of HCC and 84.4% (2 7/32) of para-carcinoma tissues. The expression of insulin-like growth factor I (IGF-I) and IFG-I receptors (IGF) was detected in HCC tissues. The positive rates of -IR) were 4 0 6% (13/32) and 5 6 3% (18/32), respectively; the positive rate of IGF-I and IGF-IR expression in the adjacent liver tissues was 500% ( 16/32) and 656% (2 1/32). The positive rate of insulin-like growth factor II (IGF-II) and IGF-II receptor (IGF-IIR) expression in hepatocellular carcinoma tissues was 78 1% (2 5 /32); the expression of IGF-II and IGF in para-carcinoma tissues was observed. The positive rate of IIR was 81 3% (2 6/32). In the HBV-DNA integration group, the positive rates of IGF-II and IGF-IIR expression in liver cancer and adjacent liver tissues were found.
