为满足水稻SSR分析需要大规模DNA样品的要求,以水稻幼苗叶片为材料,采用改良的CTAB法提取水稻基因组DNA。经质量和纯度检测结果显示:改进的方法程序简单,药品用量少,省时省力,且提取DNA的OD260/OD280均在1.8~1.9之间,琼脂糖凝胶电泳和PCR扩增条带清晰、无污染、无降解。表明该方法适用于SSR分析的水稻基因组DNA的提取,所得DNA的质量和纯度完全满足试验要求。
In order to meet the requirement of large-scale DNA samples for SSR analysis in rice, the rice seedling leaves were used as materials and the genomic DNA of rice was extracted by improved CTAB method. The results of quality and purity test showed that the improved method has the advantages of simple procedure, less dosage, time-saving and labor-saving, and OD260 / OD280 of DNA extraction ranged from 1.8 to 1.9. The agarose gel electrophoresis and PCR amplification bands were clear , No pollution, no degradation. The results showed that this method was suitable for the extraction of rice genomic DNA for SSR analysis. The quality and purity of the obtained DNA fully satisfied the experimental requirements.
