目的检测整合素αv及基质金属蛋白酶(MMP-2)和血管内皮生长因子(VEGF)在人卵巢上皮性癌的表达,探讨整合素与卵巢上皮性癌血管生成及浸润转移的关系。方法采用免疫组织化学SABC法检测60例卵巢上皮性癌组织及30例卵巢良性肿瘤中整合素αv、VEGF、MMP-2的表达。结果 60例卵巢上皮性癌中整合素αv、VEGF、MMP-2的阳性表达率分别为68.33%、73.33%和65.00%,30例卵巢良性肿瘤中三者的阳性表达率分别为13.33%、50.00%、43.33%,卵巢上皮性癌与卵巢良性肿瘤组织间3种蛋白阳性表达率差异均有统计学意义(P<0.05,P<0.01,P<0.01);整合素αv、VEGF、MMP-2与卵巢上皮性癌的分期、分级相关;整合素αv与VEGF及MMP-2的阳性表达均成正相关。结论整合素αv在卵巢上皮性癌中高表达,与临床分期及分级相关;与MMP-2及VEGF的表达呈正相关,表明整合素αv促进卵巢上皮性癌的侵袭转移。
Objective To investigate the expression of integrin αv and matrix metalloproteinase (MMP-2) and vascular endothelial growth factor (VEGF) in human ovarian epithelial carcinoma and to investigate the relationship between integrins and angiogenesis, invasion and metastasis of epithelial ovarian cancer. Methods Immunohistochemical SABC method was used to detect the expression of integrin αv, VEGF and MMP-2 in 60 cases of ovarian epithelial carcinoma and 30 cases of benign ovarian tumors. Results The positive rates of integrin αv, VEGF and MMP-2 in 60 cases of ovarian epithelial carcinoma were 68.33%, 73.33% and 65.00%, respectively. The positive expression rates of the three in 30 cases of ovarian benign tumors were 13.33% and 50.00 %, 43.33%, respectively. There were significant differences in the positive expression rates of three proteins between epithelial ovarian cancer and benign ovarian tumor (P <0.05, P <0.01, P <0.01) And ovarian epithelial cancer stage, grade related; integrin αv and VEGF and MMP-2 positive expression were positively correlated. Conclusions Integrin αv is highly expressed in epithelial ovarian cancer, which is correlated with clinical stage and grade. It is positively correlated with the expression of MMP-2 and VEGF, indicating that integrin αv promotes the invasion and metastasis of ovarian epithelial carcinoma.
