目的:建立高效液相色谱法测定穿心莲丸中穿心莲内酯和脱水穿心莲内酯含量的方法。方法:以Agilent TC-C18柱(5μm,4.6mm×250mm)为色谱柱,以甲醇-水(55:45)为流动相,流速1.0mL/min,检测波长为245nm。结果:该方法线性关系良好,线性范围为穿心莲内酯0.0224~0.2240μg,脱水穿心莲内酯0.04~0.40μg。平均加样回收率(n=3)分别为97.51%(RSD=1.11%)和97.76%(RSD=1.31%)。结论:高效液相色谱法分离度良好,结果准确可靠,为控制穿心莲丸质量提供了一定的依据。
Objective: To establish a method for determination of andrographolide and dehydroandrographolide in Andrographis Paniculata by HPLC. METHODS: The mobile phase consisted of Agilent TC-C18 column (5μm, 4.6mm × 250mm) with methanol-water (55:45) as mobile phase at a flow rate of 1.0mL / min and a detection wavelength of 245nm. Results: The linear relationship between the method was good, the linear range of andrographolide 0.0224 ~ 0.2240μg, dehydroandrographolide 0.04 ~ 0.40μg. The average recoveries (n = 3) were 97.51% (RSD = 1.11%) and 97.76% (RSD = 1.31%), respectively. Conclusion: HPLC has good resolution, accurate and reliable results and provides a basis for the control of Andrographis paniculata.
