本研究以杂交狼尾草(Pennisetum americanum×P.purureum)种子和心叶为外植体研究不同质量浓度2,4-D与6-BA组合对愈伤组织诱导及6-BA,NAA对分化的影响。结果表明种子最佳的胚性诱导培养基MS+5.0mg·L-12,4-D+0.4 mg·L-1 6-BA,诱导率为75%,心叶最佳的胚性诱导培养基MS+3.0 mg·L-12,4-D+0.5mg·L-1 6-BA,诱导率为60%;种子愈伤组织诱导高于心叶,为较好外植体。种子分化的最佳培养基为MS+3mg·L-1 6-BA+0.5mg·L-1 NAA,分化率62.5%。该研究建立了稳定有效的再生体系,为杂交狼尾草的遗传转化和定向改良提供了技术支撑。
In this study, seeds and leaves of Pennisetum americanum × P.purureum were used as explants to study the effects of different concentrations of 2,4-D and 6-BA on callus induction and differentiation of 6-BA and NAA Impact. The results showed that the best embryogenic induction medium was MS + 5.0mg · L-12,4-D + 0.4 mg · L-1 6-BA with the induction rate of 75% MS + 3.0 mg · L-12,4-D + 0.5 mg · L-1 6-BA, the induction rate was 60%. The seed callus induction was higher than the heart, which was a good explant. The best medium for seed differentiation was MS + 3 mg · L-1 6-BA + 0.5 mg · L-1 NAA with a differentiation rate of 62.5%. The study established a stable and effective regeneration system, which provided technical support for the genetic transformation and directional improvement of hybrid Pennisetum.
