为准确衡量烟叶不同生长期及晾制过程中不同时期细菌数量的变化,采用8个白肋烟品种对生产过程的细菌总数进行了连续分离计数。采集烟株封顶时的顶叶、收获成熟叶片及收获调制后3周和5周的烟叶样品,经表面灭菌后,用捣碎机捣碎。然后用0.1% Nacl系列稀释,涂布于TSA培养基上,在28~30℃的培养箱内分别培养24、48、72h后进行菌落观察和计数。结果表明,不同品种的细菌菌落在颜色、形态上的差异很大,细菌数量大约在干重102~106菌落形成单位(cfu)/g。在检测的2年中,1999年的细菌总数明显低于2000年,尤其在晾制过程中这一差别更为明显。
In order to accurately measure the change of bacteria number in different growth stages and different periods during the process of tobacco drying, the total number of bacteria in the production process was continuously separated and counted by using 8 Burley tobacco cultivars. The apical leaves were collected when the tobacco plants were capped, the mature leaves were harvested, and the tobacco leaves at 3 weeks and 5 weeks after the harvest were harvested. After the surfaces were sterilized, they were mashed with a stamping machine. Then diluted with 0.1% Nacl series, coated on TSA medium, cultured in 28 ~ 30 ℃ incubator for 24,48,72 h after colony observation and counting. The results showed that the bacterial colonies of different breeds varied greatly in color and morphology with the number of bacteria being about 102-106 cfu / g dry weight. In the two years tested, the total number of bacteria in 1999 was significantly lower than in 2000, especially in the air-curing process.
