目的 探讨nm23-h1 基因对腺样囊性癌细胞分化的影响。方法 体外用阳离子脂质体介导nm23-h1质粒转染人口腔涎腺腺样囊性癌(ACC)细胞株,筛选出转染阳性细胞克隆,并植入4只棵鼠颌面部,3w后获取标本,石蜡包埋后进行常规组织学及免疫组织化学检查,实验以未转染的阴性细胞克隆植入4只裸鼠作对照。结果nm23-h1阳性ACC植入棵鼠体内后肿瘤细胞有向原代细胞回归的倾向,即由实体型向筛状型或管状型转变,肿瘤细胞周围的肿瘤间质增多,巢状的肿瘤细胞外围有基底膜样结构形成;肿瘤细胞在体内增殖的过程中,表达nm23-h1的细胞数量有逐渐减少、表达强度减弱的趋势。结论 以nm23-h1为靶基因的基因治疗因同时具有抑制转移和促进分化的作用,具有较大的研究价值和临床应用前景。
Objective To investigate the effect of nm23-h1 gene on the differentiation of adenoid cystic carcinoma cells. Methods The human adenoid cystic carcinoma (ACC) cell line was transfected with cationic liposome-mediated nm23-h1 plasmid, and the cloned positive cells were screened. The specimens were obtained and embedded in paraffin for routine histological and immunohistochemical examination. Four non-transfected negative cell clones were implanted into nude mice as controls. Results There was a tendency of the primary cells returning to tumor cells after the implantation of nm23-h1 positive ACC in vivo, that is, the transition from solid type to sieve type or tubular type, the increase of tumor interstitial around the tumor cells, the nested tumor cell periphery There is a basement membrane-like structure formation; the proliferation of tumor cells in the process, the number of cells expressing nm23-h1 gradually decreased, the expression intensity weakened. Conclusion The gene therapy targeting nm23-h1 gene has the potential of inhibiting metastasis and promoting differentiation. It has great value in research and clinical application.
