目的:探讨高脂血症大鼠炎症水平与肝组织中嘌呤受体P2X7,A2A表达的关系及怡瑞胶囊的干预作用。方法:SD雄性大鼠50只,随机分为正常组10只,喂饲维持饲料;高脂饲料组40只,喂饲高脂饲料,14 d后再分为模型组和怡瑞胶囊低、中、高剂量组(140,280,560 mg·kg-1),每组10只。ig给药30 d,采血分离血清,检测血脂水平。酶联免疫吸附试验(ELISA)检测白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)的水平,反转录PCR技术(RT-PCR)检测肝组织的嘌呤受体P2X7,A2A的表达。结果:与正常组比较,高脂血症模型组大鼠血清中炎症因子IL-1β和TNF-α含量显著升高(P<0.01),肝脏中嘌呤受体P2X7,A2A的表达水平升高(P<0.01),与模型组比较,怡瑞胶囊各剂量组IL-1β和TNF-α的水平显著降低(P<0.01),肝组织中P2X7,A2A的表达水平显著降低(P<0.01)。结论:怡瑞胶囊调节高脂血症大鼠肝组织中嘌呤受体的表达可能与其降低炎症水平和改善脂代谢有关。
Objective: To investigate the relationship between the inflammatory levels of hyperlipidemic rats and the expression of purine receptors P2X7 and A2A in liver tissue and the intervention of YiRui Capsule. Methods: Fifty male Sprague-Dawley rats were randomly divided into normal group (n = 10) and fed with maintenance diet. Forty fat-fed mice were fed with high-fat diet and then were divided into model group and Yi Rui Capsule , High-dose group (140,280,560 mg · kg-1), 10 rats in each group. ig administration 30 d, blood separation serum, serum lipid levels. The levels of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) were detected by enzyme linked immunosorbent assay (ELISA), and the purine receptor P2X7 was detected by reverse transcription polymerase chain reaction A2A expression. Results: Compared with the normal group, the levels of IL-1β and TNF-α in sera of rats with hyperlipidemia were significantly increased (P <0.01) and the levels of P2X7 and A2A in the liver were increased Compared with the model group, the levels of IL-1β and TNF-α in each dose group of Yi Rui Capsule were significantly decreased (P <0.01), and the expression levels of P2X7 and A2A in liver tissue were significantly decreased (P <0.01). Conclusion: Yi Rui Capsule regulates the expression of purinergic receptors in the liver tissue of hyperlipidemic rats may be related to its reduction of inflammation and improvement of lipid metabolism.
