利用SSR标记技术对‘冬枣’自然授粉实生苗1728株和‘灵宝大枣’自然授粉实生苗530株进行父本鉴定,利用可能的父本材料从376对引物中筛选出8对条带清晰、多态性较高且具有高度区分度的引物。采用荧光M13毛细管电泳技术,用这8对引物在亲本材料中共检测到45个多态等位条带,每对引物的平均多态条带数为5.625,PIC值变幅为0.58~0.86,平均为0.71。利用Cervus3.0软件对具有8对引物数据的子代进行父本分析,鉴定出‘冬枣’ב木枣’实生苗399株、‘冬枣’ב小芽枣’实生苗390株、‘冬枣’ב映山红’实生苗357株、‘灵宝大枣’ב紫圆枣’(‘紫枣’)实生苗204株、‘灵宝大枣’ב柿饼枣’实生苗126株。
SSR markers were used to identify 1728 natural pollination seedlings of Dongzao and 530 natural pollination seedlings of Lingbao jujube. Eight pairs of primers were screened out from 376 pairs of primers for possible clear bands , High polymorphism and highly discriminating primers. Using fluorescent M13 capillary electrophoresis, 45 polymorphic allelic bands were detected in the parent material using these 8 pairs of primers. The average number of polymorphic bands per primer pair was 5.625 and the PIC value ranged from 0.58 to 0.86. The average Is 0.71. Cervus3.0 software was used to carry out the paternal analysis of the progenies with 8 pairs of primers, and identified 399 seedlings of ’jujube’ × ’jujube’, 390 of ’jujube’ × ’buds’ and ’jujube’ ’×’ azalea ’357 seedlings,’ Lingbao jujube ’×’ purple dates’ (’purple jujube’) 204 seedlings, ’Lingbao jujube’ × ’Persimmon jujube’ 126 seedlings.
