对人原代表皮细胞在无血清培养基 A、B和 C中的生长过程进行动态观察 ,并于培养第 5 d用 MTT法和3H- Td R掺入法分别测定其生长和代谢情况。结果表明 :人原代表皮细胞在无血清培养基 A中生长 (OD值 :0 .6 38± 0 .0 33)和代谢 (COM:92 8± 46 .6 9)优于无血清培养基 B(OD值 :0 .5± 0 .0 37,CPM:740 .11± 43.2 9) ,差异有极显著意义 (MTT法 :P<0 .0 1,3H - Td R掺入法 :P<0 .0 1)。在无血清培养基 B中人原代表皮细胞在培养第 12 d仍未汇合联接成片 ;而在无血清培养基 C中细胞的生长增殖与无血清培养基 A相似 (P>0 .0 5 ) ,细胞在这二种培养基中于培养第 12 d均可汇合联接成片。
The growth of human primary epidermal cells in serum-free medium A, B and C was observed. The growth and metabolism of human primary epidermal cells were measured by MTT assay and 3H-TdR incorporation assay on day 5 of culture. The results showed that human primary epidermal cells grew better in serum-free medium A (OD: 0.638 ± 0.033) and metabolized (COM: 92 8 ± 46.69) than serum-free medium B (OD: 0 .5 ± 0. 037, CPM: 740.11 ± 43.29), the difference was significant (MTT method: P <0. .0 1). Human primary epidermal cells in serum-free medium B still did not converge into pieces at day 12; however, the growth of the cells in serum-free medium C was similar to serum-free medium A (P> 0.05 ), Cells in these two kinds of culture medium in the 12th day of culture can be joined together into pieces.
