目的:检测反转录病毒介导的脑肿瘤基因治疗过程中具有复制能力野生型反转录病毒(RCR)。方法:RCR检测主要方法有:DNA聚合酶链式反应(PCR),反转录PCR(RT/PCR),Southern杂交以及neo基因和lacZ基因的补救分析。结果:建立了RCR的检测系统,从DNA水平、RNA水平和病毒活体水平对产HSV-TK病毒包装细胞(TK/VPC),C6转TK基因细胞和实验动物血液可能存在的RCR野生型病毒进行检测,结果均没有检测到野生型反转录病毒。结论:采用的反转录病毒基因转移系统没有产生野生型病毒,为临床试验的安全进行提供了保证。
OBJECTIVE: To test for replication-competent wild type retrovirus (RCR) in retrovirus-mediated brain tumor gene therapy. Methods: The main methods of RCR detection were DNA polymerase chain reaction (PCR), reverse transcription PCR (RT / PCR), Southern hybridization and remediation analysis of neo gene and lacZ gene. RESULTS: The RCR detection system was established to detect RCR wild-type virus that may exist in the bloodstream of HSV-TK virus packaging cells (TK / VPC), C6 transgenic TK cells and experimental animals from DNA level, RNA level and viral live body level No wild-type retrovirus was detected or detected. CONCLUSION: The retroviral gene delivery system used did not produce wild-type virus and provided a guarantee for the safety of clinical trials.
