目的研究蕨菜黄酮提取物对小鼠急性乙醇肝毒性的保护作用。方法60只小鼠随机分成乙醇模型组、空白对照组和低、中、高3个黄酮剂量组。3个剂量组连续灌胃45 d蕨菜黄酮后1次灌胃50%乙醇;乙醇模型组连续灌胃45 d纯净水后灌胃一次50%乙醇。空白对照组连续灌胃45 d纯净水。测定血清中谷丙转氨酶(GPT)活性、肝脏中甘油三酯(TG)和丙二醛(MDA)的浓度及肝脏指数。结果与乙醇模型组GPT(33.1546±13.5162),TG(0.1077±0.0291)相比,低剂量蕨菜黄酮提取物可降低血清GPT(26.9727±9.8373)、肝脏TG(0.0776±0.0281)水平(P<0.05);中剂量可降低血清GPT(17.3500±1.7656)(P<0.01)、肝脏TG(0.0669±0.0264)水平(P<0.01)和肝脏MDA(0.4493±0.1501)水平(P<0.05);高剂量可降低血清GPT(30.1714±16.4587)、肝脏TG(0.0751±0.0300)水平(P<0.05)和肝脏MDA(0.3897±0.0935)水平(P<0.01);空白对照组肝脏指数最低,蕨菜黄酮提取物未使增大的肝脏指数恢复到正常水平。结论蕨菜黄酮提取物对小鼠急性乙醇肝毒性有保护作用。
Objective To study the protective effect of fern flavonoids extract on acute hepatotoxicity in mice. Methods Sixty mice were randomly divided into three groups: ethanol model group, blank control group and low, medium and high dosage of flavonoids. The three dose groups were fed with ferment flavonoids for 45 days and then with 50% ethanol once. The rats in model group were fed with 50% ethanol after 45 days of pure water. Blank control group, continuous infusion of 45 d pure water. Serum alanine aminotransferase (GPT) activity, liver triglyceride (TG) and malondialdehyde (MDA) concentrations and liver index were measured. Results Compared with the GPT group (33.1546 ± 13.5162) and TG (0.1077 ± 0.0291), the low-dose fern flavonoids extract could lower the serum GPT (26.9727 ± 9.8373) and liver TG (0.0776 ± 0.0281) (P <0.01); the levels of TG (0.0669 ± 0.0264) and MDA (0.4493 ± 0.1501) in the liver (P <0.05); the high dose (P <0.05) and the level of MDA (0.3897 ± 0.0935) in liver (P <0.01). The liver index of the blank control group was the lowest, while that of the fern flavone extract The increased liver index returned to normal levels. Conclusion Fern flavonoids extract has a protective effect on acute ethanol hepatotoxicity in mice.
