应用马来酸酐和苯乙烯聚合成共聚物,并将它配成丁酮-乙腈溶液,把共聚物固定到聚苯乙烯试管内壁上,借此将二抗IgG结合到试管壁上,制备成二抗固化管.利用该管建立了固相放免分析血浆ACTH以及双抗固相分离血清皮质醇RIA.所建方法的标准曲线、相关性、精密度、准确性及灵敏度都十分满意,与各自液相方法对照,测定病人样品,结果一致,两者相关密切.这些固相方法简化了操作步骤,便于推广应用.
Maleic anhydride and styrene were polymerized into a copolymer and dubbed as a butanone-acetonitrile solution. The copolymer was immobilized on the inner wall of a polystyrene tube to bind the secondary IgG to the wall of the test tube to prepare a Second anti-curing pipe. The tube was used to establish solid phase radioimmunoassay for plasma ACTH and double-antibody solid phase separation of serum cortisol RIA. The standard curve, correlation, precision, accuracy and sensitivity of the proposed method are very satisfactory. The results of the method are consistent with those of the liquid method and the results of the two methods are close. These solid-phase methods simplify the operation steps and are easy to popularize.
