目的 研究CagA阳性Hp培养滤液对人胃粘膜上皮细胞 (GES 1)作用的机制。方法 将CagA阳性Hp培养滤液诱导恶性转化的GES 1细胞与CagA阴性Hp培养滤液处理的GES 1细胞进行mRNA差异显示 ,以寻找Hp致胃癌发病的相关基因。结果mRNA差异显示片断之一命名为PFN1,仅在Hp(CagA+ )培养滤液处理的GES 1细胞中表达 ,斑点杂交结果与之一致。与GeneBank资料序列同源性比较分析提示PFN1与泛素活化酶E1同源性 99% ,即为人类泛素活化酶E1基因的一部分。结论 泛素 蛋白水解酶复合体通路中的泛素活化酶E1基因可能参与了Hp的CagA诱导的胃上皮细胞恶性转化过程。
Objective To investigate the mechanism of CagA-positive Hp culture filtrate on human gastric mucosal epithelial cells (GES 1). Methods The mRNA of GES 1 cells induced by CagA positive Hp culture filtrate was compared with that of GES 1 cells treated with CagA negative Hp culture filtrate to find out the Hp-related genes. Results One of the mRNA differential display fragments was named PFN1 and was expressed only in GES1 cells treated with Hp (CagA +) culture filtrate. The result of dot blot was consistent with that. The homology comparison with GeneBank data sequence suggested that PFN1 shared 99% homology with ubiquitin-activating enzyme E1, which was a part of human ubiquitin-activating enzyme E1 gene. Conclusion The ubiquitin-activating enzyme E1 gene in ubiquitin-proteasome pathway may be involved in the CagA-induced malignant transformation of gastric epithelial cells in Hp.
