目的:研究大鼠灌胃刺五加提取物后血浆和胆汁中刺五加苷D(M0)及其代谢产物。方法:大鼠灌胃给予刺五加提取物,分别采集给药后1h、1.5h、2h、4h、6h后肝门静脉血液和0~12h胆汁,采用UPLC-Q-TOF-MS技术结合Metabolynx XS软件,对大鼠血浆和胆汁中刺五加苷D及其代谢产物进行分析。结果:基于质谱裂解行为和文献报道,在大鼠血浆和胆汁中未检测到M0,但在血浆中检测到刺五加苷D代谢物(M1),在胆汁中检测到M1的进一步代谢物(M2)。其中M1和M2是作者未见文献报道的刺五加苷D的代谢产物。结论:刺五加苷D在大鼠体内是在去葡萄糖后,以葡萄糖醛酸化和去甲基化的形式被代谢。
OBJECTIVE: To study the acanthopanax D (M0) and its metabolites in plasma and bile after intragastric administration of Acanthopanax senticosus extract. Methods: The rats were given Acanthopanax senticosus extract by gavage, and the hepatic portal vein blood and 0-12h bile were collected at 1h, 1.5h, 2h, 4h and 6h after administration, and then analyzed by UPLC-Q-TOF-MS combined with Metabolynx XS Software for the analysis of acanthopanax D and its metabolites in rat plasma and bile. Results: M0 was not detected in plasma and bile of rats based on mass spectroscopic lysis behavior and in the literature, but electuary D metabolite (M1) was detected in plasma and further metabolite of M1 was detected in bile M2). Among them, M1 and M2 are the metabolites of acanthopanax D which were not reported in the literature. CONCLUSION: Acanthopanax D is metabolized in the form of glucuronidation and demethylation in rats following glucose deprivation.
