目的:研究缺血对豚鼠心室M细胞缓慢激活型延迟整流钾通道电流(Iks)的影响。方法:利用全细胞膜片钳技术观察豚鼠心室M细胞Iks,利用不同成分浴槽液模拟细胞正常及缺血环境,观察Iks尾电流锋值的变化情况。结果:指令电压≥0mV时,缺血组电流显著小于正常组,P<0.05;指令电压小于0mV时,两组间无显著性差异,P>0.05。结论:缺血时M细胞Iks显著减弱,可能会增加心室壁电活动不均一性,促使心律失常的发生.
AIM: To investigate the effect of ischemia on delayed activation potassium channel current (Iks) in guinea pig ventricular M cells. Methods: Whole-cell patch-clamp technique was used to observe the Iks of guinea pig ventricular M cells, and the normal and ischemic environments of the cells were simulated by bath compositions with different compositions. The changes of the Iks tail current fronts were observed. Results: When the command voltage was greater than or equal to 0mV, the current in ischemic group was significantly lower than that in normal group (P <0.05). When the command voltage was less than 0mV, there was no significant difference between two groups (P> 0.05). CONCLUSION: Iks in M cells are significantly decreased during ischemia, which may increase the inhomogeneity of ventricular wall electrical activity and induce arrhythmia.
