目的:考察杠柳不定根生长特性以及次生产物杠柳毒苷积累特性,并在此基础上考察Ag+,La3+2种诱导子对杠柳不定根生长与杠柳毒苷积累的影响。方法:每4 d对杠柳不定根进行取样,测量干重及杠柳毒苷含量,绘制杠柳不定根生长及杠柳毒苷积累曲线;在不定根生长稳定期加入不同浓度的诱导子Ag+,La3+以考察适宜杠柳毒苷生物合成的最佳浓度,并将此浓度的诱导子添加到杠柳不定根不同生长阶段以考察诱导子对不定根生长以及对杠柳毒苷生物合成的影响。结果:杠柳不定根生长特性呈典型S型生长曲线,其与杠柳毒苷的代谢呈半偶联关系。诱导子Ag+,La3+诱导杠柳毒苷生物合成的最适浓度均为0.05 mmol.L-1,且在指数生长期末期加入这2种诱导子,诱导杠柳毒苷生物合成效果最好。结论:杠柳不定根生长与其次生产物杠柳毒苷积累呈半偶联关系;诱导子Ag+和La3+的添加浓度与添加时间能明显的影响杠柳不定根生长与杠柳毒苷的生物合成。
OBJECTIVE: To investigate the characteristics of adventitious root growth and the accumulation of secondary periaruloside, and to study the effects of two Ag + and La3 + inductors on the adventitious root growth and the accumulation of periotropin. Methods: Adventitious roots of Periwinkle were sampled every 4 days, the dry weight and the content of perinoside were measured, and the adventitious root growth and the accumulation curve of periuidosis were drawn. Adventitious roots with different concentrations of Ag +, La3 + The optimal biosynthesis of perlithidin was investigated, and the inducer of this concentration was added to different growth stages of adventitious root to investigate the effects of elicitors on adventitious root growth and biosynthesis of perlipidein. Results: The adventitious root growth characteristics of Periploca sepium showed a typical S-shaped growth curve, which was semi-coupled with the metabolism of perlidin. The optimum concentration of inducing seeds Ag + and La3 + to induce the biosynthesis of perlithin was 0.05 mmol.L-1, and the two inducers were added at the end of exponential phase to induce the best biosynthesis of perlithin. CONCLUSION: Adventitious root growth of Periplocta dunya is semi-coupled with the accumulation of periocidoside. The concentration of Ag + and La3 + can significantly influence the adventitious root growth and the biosynthesis of peridotoxin.
