目的观察养阴通络方对脑缺血大鼠神经细胞凋亡的作用,并探讨其可能的作用机制。方法将大鼠随机分组,先用长期激怒法致肝肾阴虚证动物模型,在此基础上,再采用线栓法制作局灶性脑缺血/再灌注模型,运用TUNEL法检测各组缺血侧皮层和海马神经细胞的凋亡,免疫组化法检测GSK-3β、Caspase-3蛋白的表达。结果与模型组比较,养阴通络方组能减少TUNEL染色阳性细胞数,GSK-3β及Caspase-3表达水平明显低于模型组(P<0.01或P<0.05)。结论养阴通络方能减少大鼠脑缺血/再灌注区的神经细胞凋亡,抗凋亡的机制可能与降低GSK-3β、Caspase-3的表达有关。
Objective To observe the effect of Yangyin Tongluo Prescription on neuronal apoptosis in rats with cerebral ischemia and to explore its possible mechanism. Methods The rats were randomly divided into two groups. First, the animal model of liver-kidney-yin deficiency caused by long-term anger was established. On the basis of this, the model of focal cerebral ischemia / reperfusion was established by the method of thread embolism. Blood-brain cortex and hippocampal neuronal apoptosis were detected by immunohistochemistry. The expressions of GSK-3β and Caspase-3 protein were detected by immunohistochemistry. Results Compared with the model group, Yangyintongluo Prescription could reduce the number of TUNEL-positive cells, the expression levels of GSK-3β and Caspase-3 were significantly lower than those of model group (P <0.01 or P <0.05). Conclusion Yangyin Tongluo Recipe can reduce the apoptosis of neurons in cerebral ischemia / reperfusion area in rats. The mechanism of anti-apoptosis may be related to the decrease of the expression of GSK-3β and Caspase-3.
