以L-苯丙氨酸为配合剂,Cu ̄(2+)配合离子,建立了一种灵敏和简便的手性配合交换RP-HPLC分析肝微粒体中氧氟沙星对映体的方法。该法的线性范围为0.2~750μg/ml,检测限LOD为2μg/ml(S/N>3),平均回收率S型97.85%±1.88%(n=10),R型99.60%±2.20%(n=10)。该法有良好的重现性和精密度(RSD<10%),已用于经苯巴比妥诱导的大鼠肝微粒体中氧氟沙星对映异构体葡醛酸缀合反应的分析。
Using L-phenylalanine as complexing agent and Cu 2+ ion, we established a sensitive and simple method for the chiral ligand exchange RP-HPLC analysis of ofloxacin enantiomers in liver microsomes. The linear range of this method was 0.2-750μg / ml, the limit of detection LOD was 2μg / ml (S / N> 3), and the average recovery rate was 97.85% ± 1.88% (n = 10) Type 99.60% ± 2.20% (n = 10). This method has good reproducibility and precision (RSD <10%) and has been used for phenoxybic acid conjugation of ofloxacin enantiomer in rat liver microsomes induced by phenobarbital analysis.
