目的:探讨α-辅肌动蛋白(α-actinin)在脑缺血大鼠海马及心肌组织中表达的变化情况,并观察“脑心同治”方对其表达的影响。方法:采用大脑中动脉堵塞(MCAO)法制作大鼠脑缺血模型,随机分为假手术组、模型组、丹红组、金纳多组。分别对各组进行大鼠神经功能评分,脑梗死体积、脑水含量及血脑屏障通透性检测,并采用Western blot方法检测海马及心肌组织α-actinin蛋白表达。结果:脑缺血大鼠神经功能缺失,脑梗死率、脑水含量增加(P<0.01),血脑屏障通透性增大(P<0.01),海马及心肌组织α-actinin含量降低(P<0.01);药物干预后,神经功能评分降低,脑梗死率、脑水含量及血脑屏障通透性有明显改善(P<0.01),并对海马组织α-actinin蛋白含量有显著调整作用(P<0.05,P<0.01),另外丹红注射液对脑缺血状态下心肌组织中的α-actinin蛋白含量改变亦有纠正作用(P<0.01)。结论:α-actinin蛋白表达的降低可能参与了脑缺血大鼠血脑屏障通透性的改变,“脑心同治”方可以上调MCAO大鼠脑及心肌组织α-actinin蛋白的表达,推测α-actinin蛋白很可能是心脑血管疾病的病理基础及脑心同治应用研究方面的潜在靶点。
OBJECTIVE: To investigate the changes of α-actinin expression in hippocampus and myocardium of rats with cerebral ischemia and to observe the effect of “Naoxintongzhi” prescription on its expression. Methods: The rat model of cerebral ischemia was established by middle cerebral artery occlusion (MCAO). The rats were randomly divided into sham operation group, model group, Danhong group and Ginaton group. Neurological function score, volume of cerebral infarction, brain water content and permeability of blood-brain barrier were detected in each group. The expression of α-actinin in hippocampus and myocardium was detected by Western blot. Results: The neurological deficit, cerebral infarction rate, brain water content increased (P <0.01), blood-brain barrier permeability increased (P <0.01), and the content of α-actinin in hippocampus and myocardium decreased (P <0.01). After intervention, the scores of neurological function decreased, cerebral infarction rate, brain water content and permeability of blood-brain barrier were significantly improved (P <0.01), and the content of α-actinin in hippocampus was significantly regulated P <0.05, P <0.01). In addition, Danhong injection could also correct the change of α-actinin in myocardial tissue under cerebral ischemia (P <0.01). CONCLUSION: The decreased expression of α-actinin may be involved in the changes of the permeability of the blood-brain barrier in rats with cerebral ischemia, and “Naoxintongzhi” can up-regulate the expression of α-actinin in brain and myocardium of MCAO rats, It is speculated that α-actinin protein is likely to be the pathological basis of cardiovascular and cerebrovascular diseases and brain and heart Tongzhi application potential research.
