采用高效液相色谱法,在Chiralpak AD-H手性柱上成功拆分了DL-苏式-对甲砜基苯丝氨酸乙酯。考察了流动相中醇类极性改性剂、酸碱添加剂对手性分离的影响。以正己烷-异丙醇-乙醇-二乙胺-三氟乙酸(75:17:8:0.1:0.1,V/V)为流动相,流速1 mL/min、紫外检测波长224 nm、柱温30℃条件下,DL-苏式-对甲砜基苯丝氨酸乙酯得到良好的分离,分离因子为2.16,分离度为3.85;该方法稳定、重复性好,适用于此类手性药物的生产及质量控制。
DL-threo-p-methylsulfonylbenzene serine ethyl ester was successfully resolved on a Chiralpak AD-H chiral column by high performance liquid chromatography. The effects of alcoholic polar modifiers and acid-base additives on the chiral separation were investigated. The mobile phase was n-hexane-isopropanol-ethanol-diethylamine-trifluoroacetic acid (75:17:8: 0.1: 0.1, V / V) with a flow rate of 1 mL / min and UV detection wavelength of 224 nm. Under the condition of 30 ℃, DL-threo-p-methylsulfonylbenzene serine ethyl ester was well separated with a separation factor of 2.16 and a resolution of 3.85. The method was stable and reproducible and suitable for the production of such chiral drugs And quality control.
