目的:观察氯化钾(KCl)溶液刺激大鼠E17大脑皮质神经细胞后不同时间点BDNF的表达,初步探讨BDNF在核酸水平的表达变化特点及其在脑皮质神经细胞接受刺激后反应早期功能活动中的作用及影响。方法:原代细胞培养获取胎龄E17大鼠大脑皮质神经细胞;采用反转录-聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)技术检测BDNF在经KCl刺激0.5~24 h后的表达变化情况。结果:原代细胞培养显示细胞状态良好。RT-PCR结果显示:经KCl刺激后,各组不同时间点神经细胞均有BDNF mRNA的表达,其表达趋势为先下降后上升,在刺激后0.5 h表达低于正常对照组(P<0.05),且24 h达高峰。结论:KCl刺激可使原代培养的大鼠大脑皮质神经细胞中BDNF mRNA的表达增高,其表达与KCl刺激时间长短有关。
