目的:探讨益肾化湿颗粒对糖尿病大鼠肾组织Fox01表达的影响,探讨益肾化湿颗粒对糖尿病肾病模型大鼠肾保护作用的可能机制。方法:采用STZ诱导糖尿病肾病大鼠模型,大鼠随机分为正常组、模型组、益肾化湿治疗组、洛汀新治疗组。观察各组大鼠治疗后肾功能(BUN、Scr)、肾脏肥大指数、血β2微球蛋白含量及Fox01的表达等指标的变化。结果:与模型组比较,益肾化湿颗粒治疗组大鼠血BUN、Scr肾脏肥大指数显著改善(P<0.01),血β2-MG明显下降(P<0.01),肾小球Fox01阳性表达率明显下降(P<0.01),益肾化湿颗粒组与洛汀新组比较,各项指标无显著性差异(P>0.05)。结论:益肾化湿颗粒可能通过降低肾小球Fox01的表达,使肾小球细胞凋亡延缓,从而可以达到保护肾脏,改善肾脏功能的目的。
Objective: To investigate the effects of Yishen Huashi granule on the expression of Fox01 in the kidney of diabetic rats, and to explore the possible mechanism of Yishen Huashi granule on renal protective effect of diabetic nephropathy rats. Methods: The STZ-induced diabetic nephropathy rat model was established. The rats were randomly divided into normal group, model group, YISHENHUA treatment group and Lotensin treatment group. The changes of renal function (BUN, Scr), renal hypertrophy index, blood β2 microglobulin content and the expression of Fox01 in each group were observed. Results: Compared with the model group, the renal hypertrophy index of BUN and Scr in Yishen Huatan Granules treatment group was significantly improved (P <0.01), the level of β2-MG was significantly decreased (P <0.01), the expression of Fox01 in glomerulus (P <0.01). There was no significant difference in each index between Yishen Huatan Granules group and Lotensin group (P> 0.05). Conclusion: Yishen Huashui Granule can delay the apoptosis of glomerular cells by decreasing the expression of FoxO1, which can protect the kidneys and improve renal function.
