目的探讨白细胞介素10(IL-10)的表达和调控,在角质形成细胞诱导免疫抑制作用中的意义。方法从体外建立的混合表皮淋巴细胞培养体系(MELC)中,分离贴壁的角质形成细胞,并分析IL-10的表达来源。我们将抗IL-10单抗引入培养体系中,观察其对角质形成细胞所诱导的免疫抑制的影响。结果在混合培养体系中,IL-10主要来源于角质形成细胞。抗IL-10单抗可有效地阻断角质形成细胞所诱导的免疫抑制效应。结论IL-10的表达在角质形成细胞所诱导的免疫抑制效应中可能起重要作用。
Objective To investigate the expression and regulation of interleukin-10 (IL-10) and its significance in the induction of immunosuppression by keratinocytes. Methods Adherent keratinocytes were isolated from mixed cutaneous lymphocyte culture system (MELC) established in vitro and the expression of IL-10 was analyzed. We introduced anti-IL-10 mAb into the culture system and observed its effect on keratinocyte-induced immunosuppression. Results In a mixed culture system, IL-10 mainly derived from keratinocytes. Anti-IL-10 mAb is effective in blocking the immunosuppressive effects induced by keratinocytes. Conclusion IL-10 expression may play an important role in the immunosuppressive effects induced by keratinocytes.
