目的:建立可以同时测定金柴胶囊中绿原酸和黄芩苷含量的变波长HPLC法。方法:采用HPLC法,色谱柱为Cosmosil C_(18)柱(250mm×4.6mm,5μm);流动相为乙腈-0.1%磷酸水溶液,梯度洗脱;流速为1.0mL/min;柱温为30℃;波长:绿原酸(327nm)和黄芩苷(280nm)。结果:绿原酸和黄芩苷均获得良好的分离度,阴性样品不干扰样品的测定;绿原酸和黄芩苷分别在13.3~53.2μg/mL和55~220μg/mL进样浓度范围内与峰面积呈良好的线性关系。绿原酸和黄芩苷的平均回收率分别为99.4%(RSD=1.6%)和99.8%(RSD=0.9%)。结论:该方法简便、准确、重复性好、专属性强,可用于金柴胶囊的质量控制和评价。
OBJECTIVE: To establish a variable wavelength HPLC method for the simultaneous determination of chlorogenic acid and baicalin in Jinchai Capsules. Methods: The chromatographic column was Cosmosil C_ (18) column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-0.1% phosphoric acid solution with gradient elution at a flow rate of 1.0 mL / ; Wavelength: chlorogenic acid (327 nm) and baicalin (280 nm). Results: Chlorogenic acid and baicalin all obtained good resolution. Negative samples did not interfere with the determination of the samples. Chlorogenic acid and baicalin were peaked at 13.3-53.2μg / mL and 55-220μg / mL respectively Area showed a good linear relationship. The average recoveries of chlorogenic acid and baicalin were 99.4% (RSD = 1.6%) and 99.8% (RSD = 0.9%), respectively. Conclusion: The method is simple, accurate, reproducible and specific. It can be used for the quality control and evaluation of Jinchai Capsules.
