应用柑桔黄龙病单克隆抗体,通过胶体金标记,检测病叶徒手切片,在400 ×显微镜下观察,韧皮部病灶呈黑褐色斑点;用异硫氰荧光素标记二次抗体法检测病叶切片,在激光共焦扫描显微镜下观察,病叶韧皮部呈现白色荧光斑点.经扫描其特异荧光强度平均值,可作定量参考.按 Jagoueix 报道的黄龙病病原16s D N A 序列,设计引物,用 P C R 技术检测病叶 D N A,获得特异性扩增区带,约1160bp ,可作为诊断依据.
