目的分析三结构域蛋白28(TRIM28)在胃癌中的表达情况及TRIM28 siRNA转染胃癌细胞后对胃癌细胞的增殖和凋亡的影响。方法培养胃癌细胞BGC-823和SGC-7901并收集30例胃癌组织与癌旁正常组织,运用Western blot方法检测胃癌细胞株和组织中TRIM28的表达;用TRIM28 siRNA转染胃癌细胞株BGC-823后培养12h,检测转染后的TRIM28的蛋白表达水平,并用MTT法和流式细胞仪检测胃癌细胞的增殖率和凋亡率。结果 TRIM28在两种胃癌细胞株中均有表达,40例胃癌组织中有27例患者的TRIM28在胃癌组织中的表达水平显著高于癌旁正常组织;运用TRIM28 si RNA转染胃癌细胞株后,TRIM28的蛋白表达水平明显下降,转染TRIM28 siRNA后的胃癌细胞的增殖率为(53.86±0.85)%,与对照组si RNA的增殖率(97.45±0.50)%比较,差异有统计学意义(P<0.05);转染TRIM28 siRNA后的胃癌细胞的凋亡率为(32.77±0.73)%,与对照组si RNA的凋亡率(4.81±0.26)%比较,差异有统计学意义(P<0.05)。结论 TRIM28在胃癌中呈现高表达,且在胃癌细胞的增殖和凋亡中发挥重要作用,TRIM28有可能成为胃癌治疗的新靶点。
Objective To analyze the expression of tridomain protein 28 (TRIM28) in gastric cancer and the effect of TRIM28 siRNA transfected gastric cancer cells on the proliferation and apoptosis of gastric cancer cells. Methods The gastric cancer cells BGC-823 and SGC-7901 were cultured and 30 cases of gastric cancer tissues and adjacent normal tissues were collected. The expression of TRIM28 in gastric cancer cell lines and tissues was detected by Western blot. After transfection of gastric cancer cell line BGC-823 with TRIM28 siRNA Cultured for 12h, detected the protein expression level of transfected TRIM28, and detected the proliferation rate and apoptosis rate of gastric cancer cells by MTT assay and flow cytometry. Results TRIM28 was expressed in both gastric cancer cell lines. The expression level of TRIM28 in gastric cancer tissue was significantly higher in gastric cancer tissues than that in adjacent normal tissues. Conclusion TRIM28 is highly expressed in gastric cancer and plays an important role in the proliferation and apoptosis of gastric cancer cells. TRIM28 may become a new target for the treatment of gastric cancer.
