目的探讨miR-145对宫颈癌HELA细胞侵袭转移的机制,寻求宫颈癌治疗的新途径。方法构建载体验证miR-145与靶基因MUC1的互补作用,利用稳定表达miR-145的HELA细胞,揭示miR-145与MUC1及上皮间质转化(EMT)三者之间的关系,发现宫颈癌细胞可能的侵袭转移方式。结果 miR-145模拟物与报告载体共转染293T细胞后,荧光表达量明显下降,P<0.05;miR-145模拟物与突变载体共转染293T细胞后,荧光表达量无明显改变,P>0.05。E-cad蛋白表达在miR-145未处理的各组HELA细胞中均低表达;在miR-145-CMV-RFP-HELA细胞系中E-cad的表达显著增强。结论 miR-145在宫颈癌中起到抑癌基因的作用,miR-145对宫颈癌细胞的侵袭转移存在负性调控作用,miR-145通过MUC1作用于EMT。
Objective To investigate the mechanism of miR-145 in invasion and metastasis of cervical cancer HELA cells and to find a new way to treat cervical cancer. Methods The constructed vector was used to verify the complementarity of miR-145 with MUC1. Using HELA cells stably expressing miR-145 to reveal the relationship between miR-145 and MUC1 and epithelial-mesenchymal transition (EMT), we found that cervical cancer cells Possible invasion and metastasis. Results After co-transfected 293T cells with miR-145 mimics and the reporter vector, the expression of miR-145 mimics and the reporter vector significantly decreased (P <0.05). There was no significant difference in the expression of miR-145 mimics and the mutant vector when transfected into 293T cells, 0.05. The expression of E-cad protein in HELA cells of miR-145 untreated group was low, and the expression of E-cad in miR-145-CMV-RFP-HELA cell line was significantly increased. Conclusions miR-145 plays a role of tumor suppressor gene in cervical cancer. MiR-145 negatively regulates the invasion and metastasis of cervical cancer cells, and miR-145 acts on EMT through MUC1.
